History:

Vardenafil was co-marketed by Bayer Pharmaceuticals, GlaxoSmithKline, and Schering-Plough under the trade name Levitra. As of 2005, the co-promotion rights of GSK on Levitra have been returned to Bayer in many markets outside the U.S. In Italy, Bayer sells vardenafil as Levitra and GSK sells it as Vivanza. Thus, because of European Union trade rules, parallel imports might result in Vivanza sold next to Levitra in the EU.

 

An orally disintegrating form, marketed as Staxyn and Levitra Soft. has been gaining approvals in countries such as the United States and Canada.

Treatment of ED:

How Does Vardenafil Work?

It has a mechanism of action that is similar to sildenafil (Viagra), and tadalafil (Cialis). Penile erection is caused by the engorgement of the penis with blood. This engorgement occurs when the blood vessels delivering blood to the penis increase in size and increase the delivery of blood to the penis. At the same time, the blood vessels carrying blood away from the penis decrease in size and decrease the removal of blood from the penis. Sexual stimulation that leads to the engorgement and erection causes the production and release of nitric oxide in the penis. Nitric oxide then activates the enzyme, guanylate cyclase to produce cyclic guanosine monophosphate (cGMP). The cGMP is primarily responsible for increasing and decreasing the size of the blood vessels carrying blood to and from the penis, respectively. Vardenafil prevents an enzyme called phosphodiesterase-5 (PDE-5) from destroying cGMP so that cGMP persists longer. The longer cGMP persists, the more prolonged the engorgement of the penis. Vardenafil was approved by the FDA in August 2003.

 

 

Contraindications:

This medication is a highly effective treatment for erectile dysfunction (impotence) in men. However, it is not meant for everyone. Here are a few warnings to consider before taking Levitra:

 

Do not take this drug if you are being treated with other medications that contain nitrates
Do not use this medication if you suffer from retinitis pigmentosa
Avoid this product it if you are allergic to vardenafil or any other ingredients in this medicine
Do not use if you have a history of severe heart, kidney and liver problems
Do not take this medicine if you are taking drugs for HIV or fungal infections
Avoid if you have non-arteritic anterior ischaemic optic neuropathy (NAION)
Do not use if you have a condition that causes bleeding such as a stomach ulcer
Do not consume if you have sickle cell disease, bone cancer or leukaemia.

 

 

Possible Side Effects:

Levitra is not known to have any serious side effects and, as a matter of fact, they probably exhibit the least negative side effects when compared to any other erectile dysfunction treatment. Mild side effects may include dizziness, facial flushing, blocked nose, nausea, impaired vision and upset stomach. If experienced, these side effects should subside within a few hours. Major side effects such as erections that last more than 4 hours, temporary vision loss or impairment of hearing are very rare. If you ever experience such effects, you need to report these immediately to a doctor.

 

 

Dosage:

Take vardenafil exactly as your doctor tells you to. It is intended to be taken before sexual activity - it is NOT for continuous daily use. You should take one tablet 25-60 minutes before you plan to have sex, although it may still be effective up to 4-5 hours after taking the tablet. You can take the tablet either before or after food but, if you have just eaten a large or high-fat meal, it may take longer for it to work.

There are three strengths of vardenafil tablets: 5 mg, 10 mg and 20 mg. It is likely that you will be given the 10 mg strength tablet to begin with, but this may be changed subsequently depending upon how you react. Each time you collect a prescription, check to make sure it contains the strength of tablet you are expecting.

If you have been given orodispersible tablets, place one tablet on your tongue and allow it to dissolve before you swallow. Do not take the tablet with anything to drink.

Do not take more than one dose of vardenafil in any 24-hour period.

 

Individuals 65 years of age or older should begin therapy with 5 mg.
